Der bescheidene Farmer Dan Evans (Van Heflin) wird zusammen mit seinen Söhnen Zeuge, wie bei einem Postkutschenüberfall der Räuberbandenboss Ben Wade (Glenn Ford) eiskalt den Kutscher erschießt. Wenig später wird Wade gefangen genommen, während dem Rest der Bande unter Leitung seiner nicht minder skrupellosen Nummer 2, Charlie Prince (Richard Jaeckel), die Flucht gelingt. Der Marshal von Bisbee (Ford Rainey) will den Gefangenen nach Contention City bringen, von wo aus der Zug um 3:10 Uhr Richtung Yuma und dem dortigen Gefängnis losfahren soll. In der Gewissheit um die Gefährlichkeit der Reise lobt Zuglininienbesitzer Mr. Butterfield (Robert Emhardt) eine Prämie von 200 Dollar für die Gewährleistung des erfolgreichen Eskorts aus. Gegen den Rat seiner Frau Alice (Leora Dana) nimmt er das Angebot an, wie auch der stadtbekannte Trinker Alex Potter (Henry Jones).
